About Semaglutide dosing

Semaglutide is a GLP-1 receptor agonist taken once a week. It slows gastric emptying and acts on appetite signalling in the brain, which is why the dose is increased gradually — the escalation schedule is what keeps nausea manageable.

A 5 mg vial in 2 mL is 2.5 mg per mL, so the 0.25 mg starting dose is 0.1 mL — 10 units. At the 2.4 mg maintenance dose the same vial gives roughly two doses, which is why larger vials are common later in treatment.

Semaglutide is a prescription medication. The dose, the escalation pace, and whether it is appropriate for you are decisions for a licensed provider; this calculator only turns a prescribed dose into a syringe measurement.

How do I reconstitute compounded semaglutide?

Add the bacteriostatic water your pharmacy specifies slowly down the side of the vial, swirl until clear, and refrigerate. 2 mL for a 5 mg vial (2.5 mg/mL) keeps the early doses at easy-to-read unit counts; enter your own volume above.

What is the typical starting dose for semaglutide?

The labelled schedule starts at 0.25 mg once weekly for four weeks, then 0.5 mg, 1 mg, 1.7 mg, and 2.4 mg, each step usually held for at least four weeks. Compounded protocols vary; your provider sets your schedule.

How long does reconstituted semaglutide last?

Follow your pharmacy's beyond-use date, commonly 28 days refrigerated. Because doses climb, a 5 mg vial covers many weeks at 0.25 mg but only about two doses at 2.4 mg, so plan refills around the step you are on.
